Staining for evaluation of estrus by vaginal cytology.
Diff Quick (3 solutions x 250 ml).
Simple staining method consisting of an eosinophilic, a basophilic and a fixative stain. The staining sequence includes 5-10 immersions in the red stain, then in the navy blue stain, followed by the light blue stain and finally with immersions in water collected from the tap if it has potable properties.
Depending on the cells that we visualize under the microscope, we will know in which stage of the estrous cycle the female is in.
FOR 500 TESTS
Kit of three colorants.
A.- Fixing solution.
B.- Acid staining.
C.- Basic staining.
Technical:
1º.- With the help of the swab, deposit a layer of cells on the slide.
2º.- Immerse the slide in the fixative solution for 5 seconds.
3º.- Place the slide on absorbent paper to remove any remaining fixative solution.
4º.- Immerse in the acid solution (red) for 5 seconds. Remove excess dye with absorbent paper.
5º.- Immerse another 5 seconds in the basic solution (navy blue).
6º.- Wash with water.
